If I have a skeleton definition for triggerProfileUpdate() and another for triggerFieldMyFieldUpdate(), do they both always fire on an update to MyField? If so, in what order?

Yes

Easy Profile trigger these with following order:
1. triggerFieldMyFieldUpdate
2. triggerProfileUpdate

So, as long as the triggerProfileUpdate() does not overlap updating the same Profile Fields as triggerFieldMyFieldUpdate() I should be fine to use both?

Which is more efficient? Should I put all my update code in triggerProfileUpdate() since I have that already set up of multiple field updates, or is there any advantage to using both?

It might be a good idea for you to add this information to your Skeleton document page, and to comments in the skeleton.php. Just a suggestion...
